چکیده

1. Extensive experimental data have been collected on the time required for the excystment process of the small ciliate Colpoda duodenaria throughout a range of temperatures of 8 degrees through 32 degrees C. and a range of concentrations of yeast extract excystment media of 0.08 through 22.4 gm./liter. 2. The excystment process has been separated into two periods, the first inversely proportional to the concentration of the yeast extract and the second independent of its concentration. 3. The first excystment period has been found to depend on the time for diffusion through the protoplasm of a compound from the yeast extract and on the time for a chemical reaction with the extremely high energy of activation of 220,000 calories/mole. 4. The changes in viscosity with temperature for this Colpoda, inferred from diffusion rate changes, have been found to be almost the same as those found by Heilbrunn for Amoeba dubia by the direct method of centrifuging granules. 5. The second excystment period is shown to be controlled by reactions whose apparent activation energies are 44,000 calories/mole below 15 degrees C. and 18,000 calories/mole above 15 degrees C.; above 25 degrees C. this period is independent of temperature. 6. The distribution of the log excystment times of individual organisms about the mean log excystment time is found to be independent of temperature except in the range where the reaction with highest activation energy takes a significant length of time, and to increase rapidly with decreasing temperatures in this range.

Excystment-Dependent Alteration of Protein Expression in Terrestrial Ciliate Colpoda cucullus

Protein expression during the excystment of Colpoda cucullus was studied by SDS-PAGE. The expression levels of 60-, 50- and 49-kDa proteins were markedly changed from the early to later stage of excystment. The 60-kDa protein (p60) was temporarily expressed first, and its expression was inhibited by actinomycin D. LC-MS/MS analysis showed that the amino acid sequences of p60 partially coincided...

Emergence of the Terrestrial Ciliate Colpoda cucullus from a Resting Cyst: Rupture of the Cyst Wall by Active Expansion of an Excystment Vacuole

The first sign of excysting Colpoda cucullus cells is the initiation of the pulsation of a contractile vacuole, which is then replaced by a non-pulsating vacuole (excystment vacuole) that continues to expand and finally ruptures the outermost cyst wall (ectocyst) due to inner pressure. A ciliate surrounded by flexible membranes (endocyst) thus emerges. The osmolarity of the excysting cells is e...

Morphological Changes and the Requirements for Macromolecule Synthesis during Excystment of Acanthamoeba Castellanii

Light and phase-contrast microscopic observations of excystment in Acanthamoeba castellanii have been used to classify cells in excysting populations as free trophozoites, or mature, activated, or preemergent cysts. These categories have been used to describe the kinetics of excystment. A pH of 7 and a temperature of 30 degrees C have been found to be optimal for the activation of mature cysts....

On the effect of osmotic pressure upon the excystment of the cysticercoids of Raillietina kashiwarensis.

played an important role in causing the excystment of the cysticercoids. But on the other hand, it is necessary to examine what effect is exerted by the osmotic pressure in the alimentary tract of the chicken upon the excystment. Up to the present time, as to the excystment of the tapeworm bladderworm, only the effect of the digestive juice has been examined but no study on the effect of the os...

In Vitro Excystment of the Metacercaria of Acanthoparyphium spinulosum (Trematoda: Echinostomatidae)

In vitro excystment studies on the metacercaria of Acanthoparyphium spinulosum indicated that optimal excystment occurred when the cysts were incubated in either a complete medium containing trypsin and bile salts, or bile salts alone following acid pretreatment, and the reductant (sodium dithionite) at a temperature of 42°C and a pH of 7.8. A relatively lower percentage of excystment was obtai...
